The invention provides an antiviral polypeptide. The amino acid sequence of the antiviral polypeptide is one or more of SEQ ID NO.2-5. The antiviral polypeptide provided by the invention is obtained by replacing or increasing specific amino acids and simultaneously retaining an alpha-spiral functional region for playing a role of the Cathelicidin polypeptide (SEQ ID NO.1) through modification, can enhance the expression of host I-type interferon, plays a relatively strong antiviral function at in-vivo and in-vitro levels, and has a half inhibition concentration to H1N1 virus of 2.47-8.14 mu M. The invention also provides a preparation method of the antiviral polypeptide. The antiviral polypeptide provided by the invention has low toxicity to cells, and can be applied to preparation of drugs or reagents for preventing and treating influenza A.
